Description
1% aqueous solution of crystal violet preparation for microbiological diagnostics, chemical reagent
Composition:
Pyoctaninum coeruleum, ethyl alcohol, auxiliary substances.
1. Microbiological staining by Gram technique:
I. Primary staining with Pyoctanine (crystal violet) solution. All bacteria are stained purple under the influence of this basic dye.
II. Fixation with Lugol (aqueous solution of iodine in potassium iodide). Iodine combined with crystal violet forms crystal-violet-iodine complexes in cells.
III. Discoloration (ethyl alcohol or a mixture of ethyl alcohol-acetone). Primary staining with crystal violet is washed out in some bacteria, others remain stained.
IV. Staining with contrast dye (carbolic fuchsine, safranin). This basic dye re-stains red (safranin) or pink (fuchsine) bacteria without colour during decolourisation.
2. Detection of fingerprints:
Immerse the test surfaces in the solution and rinse under running water. When working with the solution, be very careful, because violet is very difficult to wash with dye and may leave permanent stains on the clothes, furniture or floor.
3. Ingredient of microbial substrates:
Crystal violet is a substrate component: MacConkey Agar with violet crystal, Violet Red Bile Glucose Agar, MacConkey Agar with sorbitol, Violet Red Bile Lactose Agar, Yersinia CIN Agar, Columbia Agar with violet crystal and 5% sheep blood, Uragar, a dye for the Gram method. The preparation is not approved for use in living animals.
