Description

iodine spirit solution in potassium iodide preparation for microbiological diagnostics, chemical reagent

Composition:
iodine, potassium iodide, water, auxiliary substances.

1. Microbiological staining by Gram technique:
I. Primary staining with Pyoctanine (crystal violet) solution. All bacteria are stained purple under the influence of this basic dye.
II. Consolidation with Iodine (iodine spirit solution in potassium iodide). Iodine combined with crystal violet forms crystal-violet-iodine complexes in cells.
III. Discoloration (ethyl alcohol or a mixture of ethyl alcohol-acetone). Primary staining with crystal violet is washed out in some bacteria, others remain stained.
IV. Staining with contrast dye (carbolic fuchsine, safranin). This basic dye re-stains red (safranin) or pink (fuchsine) bacteria without colour during decolourisation.

2. Detection of honey adulterations with starch and potato syrup:
Detection of starch in honey. About 5 g of honey is diluted with a double amount of water and heated to boiling. Then, 2–3 drops of iodine (iodine spirit solution in potassium iodide) are added to the cooled solution and shaken vigorously. If the solution turns blue, it is evidence of honey starch falsification. When heating, the colour disappears, so make sure that the solution is sufficiently cooled.

3. Removal and reduction of visible scratches on wooden surfaces:
The scratched surfaces should be painted over with a mixture of vinegar and iodine (for clear furniture use more vinegar, darker ones – more iodine). The preparation is not approved for use in living animals.
